Instructions:

1. Install the supplied pressure reducing valve on the CO2 cylinder. There must be no air leakage at the joint. Do not open the cylinder temporarily. Connect the pressure reducing valve output connector with the hose on the street of the CO2 intake pipe on the back of the CO2 box, and press it with a compression ring to prevent air leakage.

2. Wipe the CO2 box working chamber with alcohol, turn on the UV lamp power switch on the back of the box, and disinfect for 1-2h.

3. Connect the faucet and the upper left side of the tank with a water pipe. The water jacketed CO2 tank operates in one operation.

4. Turn on the power, set the power switch to "1", and the green indicator light.

The water jacketed CO2 tank must first be filled with water. Open the faucet, slowly add water to the sleeping village, and gradually increase with the water level. After the local water level indicator is off, wait for 3-5s at this time, then stop adding water. At this time, the water level is between low and high, and the water level light is not bright. The equipment can be put into operation.

Adjust the temperature setting to the desired temperature (for example, 37 degrees), and the heating indicator light (green) should be on, indicating that it is heating. (Note: Ambient temperature exceeds 25 degrees, air conditioning can be used to reduce ambient temperature)

Set the CO2 control switch to "0".

After the temperature reaches the set value, the CO2 cylinder is opened. (Before opening, the pressure reducing valve should be loosened as much as possible to prevent the pressure of the pressure reducing valve from being too high, causing the rubber tube to burst!) The pressure gauge on the pressure reducing valve indicates the inside of the cylinder. CO2 pressure, slowly turn the relief valve knob clockwise. The pressure output of the pressure reducing valve is indicated as 0.05 MPa, and the pointer is in the middle of the scale line. At this time, use the dial to set the CO2 concentration (factory adjusted to 5%). Before turning on the CO2 control switch, turn off the air flow meter and CO2 flow meter. The CO2 controller displays “1”, and CO2 enters the inner chamber. As the concentration increases, the CO2 controller displays the CO2 concentration value. When the set value is reached, the device automatically switches to the air and CO2 qi state. At this time, the air flow meter should be adjusted at 760ml/min (already adjusted at the factory), the CO2 flowmeter float should be indicated at 40ml/min (already adjusted at the factory); if the float deviates from the above value, the flowmeter can be adjusted. Turn the knob to 760ml/min (air) and 40ml/min (CO2) accurately.

When the temperature reaches the set value, the fluctuation is within ±0.5 °C, and the CO2 concentration is also reached, the cell culture can be performed. Put in a water tray to make the humidity meet the requirements.

The machine should be used in accordance with the above requirements after using the machine for the first time or after long-term use, and should be used for contamination inspection inside the box before formal cultivation.

5. Shutdown

After the cultivation is completed, the culture dish is taken out of the box and the uncultured dish is re-introduced (note whether the distilled water in the water tray needs to be replenished). If the culture is stopped, the CO2 cylinder should be closed first, then the air flow meter and CO2 flow meter should be closed, and the CO2 intake switch should be set to “0”. Location, and finally turn off the main power. If it is not used for a long time, the water tray should be taken out and the working chamber should be dried. When the CO2 air inlet switch is turned off, the CO2 box is turned on at 37 °C for 2 hours, the studio is dried, and the total power is cut off.

6. Temperature adjustment method

When the machine is shipped from the factory, it is adjusted at 37 °C. For example, if the water mist on the glass door in the winter box is large, the culture in the box cannot be seen. The door temperature knob can be adjusted behind the CO2 box, and the temperature is raised to the left. In the high direction, until you can see the contents of the box; if the actual temperature in the box is higher than the set temperature in summer, turn the adjustment knob to the right, that is, the temperature will decrease until the temperature in the box reaches 37 °C.

7. Fan adjustment method

In use, the amount of air volume will not only affect the uniformity of the temperature inside the box, but also affect the control temperature. If the air volume is too large to affect the cultured item during use, adjust the fan speed knob on the back of the CO2 box until it meets the needs.

Precautions

1. This machine should be installed in a room with clean air, no direct sunlight, no strong electromagnetic field and radiant energy, and the surrounding temperature difference is relatively small. In order to ensure the temperature control accuracy of the CO2 box, it is recommended to use it in the environment of 15 °C - 25 °C.

2. Pay special attention to the cylinder before opening, you must loosen the pressure reducing valve to prevent the gas hose from blasting.

3. When the difference between the ambient temperature and the set temperature is less than (RT+5°C), the air conditioner is used to reduce the ambient temperature. During the whole process of the culture, there should be no obvious change in the ambient temperature, otherwise the ambient temperature will change. The temperature control in the CO2 box is not allowed.

4. During the use, the output pressure of the pressure reducing valve and the flow rate of the two flow meters should be monitored frequently. Do not arbitrarily twist the various valves in the control box. If there is a deviation in the concentration of the CO2 tank, you should first find out the reasons for the inaccuracy of the relevant values ​​before you can adjust it slightly.

5. When the cylinder pressure is less than 1MPa, it should be replaced in time. When replacing the cylinder, the cylinder switch should be closed first, the pressure reducing valve screw shaft should be loosened, and the pressure reducing valve should be removed and reinstalled on the new cylinder.

6. When the inner door of the CO2 box is jammed, the CO2 control switch should be turned off first. After the door is closed, when the CO2 concentration display shows “0.0”, the CO2 control switch is turned on again.
